If I remove my company domain from 'Approve the following domains' section in Site Access will this prevent users in my company being able to create their own accounts?
I want staff to be able to view our Confluence site but I don't want anyone being able to set up their own accounts so basically I don't want them all to be anonymous users.
I want to check though that this change will not affect our Admin users - will they still have full access to the site and be able to create new and edit existing content?

anonymous access has to do with public access (attention this also means it's available for everyone!)
Creating own accounts would be an admin right - you have to manage your permissions carefully. Please see this description about permissions and restrictions in Confluence Cloud here.
