As @Mykenna Cepek said It would count single license per user regardless of of how many groups you can create and add user to the group. 

for example you can add "A" user in multiple groups.

No. One license is used for each ACTIVE user in your instance, regardless of how many groups or teams or roles that user is configured for.

Note that you can deactivate users (without deleting them) to bring your licensed user count down. Deactivating them leaves their data in the system, without costing you a license seat.
