In a Confluence clustered configuration (v 5.4.4 as described here https://confluence.atlassian.com/display/CONF54/Confluence+Clustering+Overview) we want to get an XML backup.
We are planning to access a node directly using the node hostname but we are not sure the backup location is configured to a specific node somewhere in the configuration.
Is there a way to control in what node the backup is going to be stored?

In clustered Confluence, the backup is is available in the backups directory of the home directory of whichever node you are connecting to. It is not possible to configure it to go to a specific node.
I wanted to let you know as well that your Confluence version of 5.4.4 reached End of Life on Mar 18, 2016. It is at very high risk of being compromised, due to the following security advisories, explained below:
You'll need to make sure that you upgrade as soon as possible to the latest version of Confluence. For details on how to do that, have a look at our Confluence Installation and Upgrade Guide.
My recommendation would be to obtain a Data Center license for Confluence and upgrade to Confluence 5.10. Then, you can migrate to Confluence Data Center. Once this is complete, then you want to immediately move to the latest version of Confluence.
It's very important that the upgrade is completed, as we are already seeing many users who have been compromised as a result of these vulnerabilities.

What about when connecting directly to a node bypassing the load balancer?

In that case, it's in the home directory of whichever node you are connecting to.
