Does Confluence has lock file as in JIRA?

Do you mean a lock file like one that's created once the Tomcat process is running?
If so, then yes. There should be a .pid file under the "work" directory.
But our Confluence doesn't hahe one in working directory.
Do you know how to activate it?

You can set CATALINA_ID in bin/setenv.sh although I suspect it may already be set. It will take effect when you restart Confluence
