We have purchased a licence with 25 users, but we are also using Microsoft AD as user directory with local groups in Confluence.
How do I manage Confluence users from Confluence admin page?
When I open a particular users profile, there is no check box to activate/disable that account.

this is more a topic of "what users are synchronized from my AD to Confluence". It's not possible to enable/disable accounts, only to control what users are synchronized from AD.
If you have a 25 user license, you must make sure that only 25 users are synchronized from AD to Confluence. This can be achieved by writing an LDAP filter and set it at ConfluenceAdmin > User directories in the configuration of your user directory.
