We are receiving an error, page not loading properly. Error# Unknown macro: {User layout} ()
I have a production and a Dev environment. The issue is with the Dev environment. On checking the Dev, we can see that a panel Theme Builder is not there. Where as it is there in the prod. Please find the screen shot. Kindly advice if this is the issue. Also, on another pages we can see that another issue appears as Unknown macro: {align}.
Please advice...
Also, when we checked the System Information we do not find any Plugin named Theme Builder.
The version we are using is 3.4.6

Indeed you need to install the theme builder plugin. You can either install it via Confluence Admin > Plugins (or Find New Add-ons), or you can download the jar file here and upload it manually.
The "Unknown macro: {align}" is also a third party plugin, more precisely the Content Formatting plugin. You will also need to install this one in order to get rid of the error.

It seems that this page uses a macro which no longer exists. The {use-layout} macro is provided by Theme bulder plugin. Is this plugin installed and enableld in your Confluence instance? Is the plugin version compatible with your Confluence version? Is the plugin properly licensed?
