When you select "A user is defined list", you get option to add users to the list. See the screenshot.
Select "Method to choose assignee" as required for the project.

If its a JQL condition, then create 3 different rules for each groups.

- We have 3 shifts in our team
- Each Shift we have 2 or 3 members
- Weekend only 2 members will work on shifts
- Since i am new to JIRA Admin, Could you pls share any steps or screenshot or link
- I need to create the Groups naming as APJ Service Desk, EMEA Service Desk or AMR Service Desk or Weekend Service Desk and to add the members as per shift pattern
- Condition at present, i need to assign some weekend tickets according to region wise members

There is no direct answer for this. I got one comment which suggests the solution. I have not tested it.
