Within scrum board a particular column "In Testing" is not visible for the users, only board admin can see it.
Please help me to resolve this puzzle.

The board filter is quite simple. Jira cloud status In testing associated with Stort issue type which included in the filter.
project = XXXX AND issuetype not in (Test, "Test Execution", "Test Plan", "Test Set") ORDER BY Rank ASC

Does the user have access to this one issue in the In Testing status outside of the board?

Yes, user has access to the issue which is In Testing status. Also, the board is shared with all the login in users.

It looks like the user's view is grouping by Assignee. Maybe that issue is not assigned? Take a look at that. Also, have the user change the grouping on the board to see if it changes. Maybe group by Status.

Board Swimlanes is set to the assignee and the story is assigned to one of user listed on the screenshot but unfortunately, the assignee is also not able to see In Testing column.
