Changes to a workflow do not take effect until you publish the draft that you are editing at which point they take effect from that point on. There is no way to apply changes to a workflow on a partial basis.
No issues will change when you add new statuses and you can add the columns to your Agile boards after you have published the workflow. If you change or delete a status you will be prompted at publication of the workflow to decide what status they should change to.
It is important to make sure that you do not leave any statuses without an outgoing transition before you publish as issues could end up stuck there.

Adding columns would only literally add columns to your board.
Your existing task will only be impacted only if you map them to the new columns. Are you doing any workflow changes or is it just board configuration?
