How do we Forbid Jira users/Project Admins from adding any users to a specific Project role (Example: Roles like 'Product owner')? In short, how do we lock a Jira project role and make it accessible only to Jira system Admin?

The only way to prevent someone from adding users to a specific project role is to not add that someone as project admin. See Managing project roles for more information.

No, the permissions are part of the core of Jira, they're not modifiable directly.
The closest you can get is to turn off the project admin rights for people and then create projects that have issues and workflows that allow normal users to do admin actions. This can very quickly snowball into huge scripting projects!
