you can add workflow validator or In the field configuration to make the label field mandatory.
Associate the field configuration to the Task Issue type for the required project.
Workflow Validator > add this validator to the task workflow

Basically, it works like this:
A Workflow contains of Steps (Status) that are connected by Transitions. For example, moving an issue from New to Open would be a transition. Transitions themselves can be configured to have Conditions, Validators and Post Functions.
@Sachin Dhamale suggested adding a Validator and I agree with that approach.
Your second screen shows a number of Workflows – you would have to edit one of them and then pick a transition to add a Validator.

Making a field required using field configuration is slightly more blunt compared to a Validator. When you make a field required using field configuration, then it's required everywhere.
