Thank you for reaching out to Community!

Checking our API documentation and also similar questions, I didn't find any API that allows setting a custom field as required.

On the UI we set this on the Field Configuration Scheme and the APIs available for that are only "GET".

For now, there is no feature request suggesting this option, so please, feel free to raise one on jira.atlassian.com (project JRACLOUD) adding more details about why this option is important for your environment.
