I need to upgrade JIRA 4.1.2 up to 5.0.2. Is it possible to do an in place upgrade directly to the new one straight from 4.1.2, or do I have to do incremental upgrades? If incremental, which releases are required in between?

You can do a direct upgrade but make sure you understand the changes in between. For example, 4.3 has changes to user diretcories.

Go through all the release notes and upgrade notes. You should be fine if you follow the docs.
