We are interested in upgrading to Jira Software Premium. We currently have service desk as well and do not want to upgrade Service desk to premium. Are you forced to upgrade Jira Service Desk to premium if you have Jira Software Premium?

Yes, the Licenses can be mixed for the Jira Cloud Application types, and each application is going to be managed separately. And some additional details on the licensing breakdown on Standard compared to Premium can be seen in:
To modify the license type go to Site administration > Billing > Manage Subscriptions and change the license type for the desired application.
also before upgrading the license you can run comparisons of the application costs on the mixed licenses using the application price calc tool at https://www.atlassian.com/software/pricing-calculator
