Most of our users have JIRA Software license only. For any service desk ticket they always have to go to Service Desk Portal.
Is it possible for them to Raise a request from project itself in JIRA? If yes, how can they see sidebar of Service Desk Project?

If a user does not have a Jira Service Desk license, then the user has access to Service Desk feature only via the Portal
