We have a requirement where we will send mail to Jira, based on the email subject (Project name specified like, eg- <Project>:<Actaul subject>) it should create a ticket in the specified project. is it possible to create a custom plugin?. we don't need to use a different email for each project.
Enterprise Mail Handler for Jira (JEMH) will allow you to do this in multiple ways.
It is a very powerful product, though a little complex to set up till you get used to it.

Is not possible out of the box since an email belong to one project. May be there is a plugin that will parse the mail subject but i don't use such plugin so i can't recommend you wich plugin to try.
