JIRA default value issue

I created a user custom filed with default user value called "Unassigned". When the field is updated with a new user, it is not showing impact in JQL sporadically. 

JQL is working fine after project reindex (FYI the project has more than 50k issues).

Did anyone face the same issue? Can someone help me with the solution?

How do you update the field value ? Is it from a Screen or a postfunction/automation ?

Did it work from the edit screen. It seems that the issue re-index is not triggered.
