I added a custom user Picker(single user) field to the ticket type Epic and added to the view screen under a tab. I have below two issues with it:
1. This field is populating under People in the view screen and not in the tab
2. I'm not able to filter the issues with this field. Even though many tickets have this filed with a user value, I'm not able to filter it for a single user.
Please help.

Allow me to bring some details about both issues you mentioned so we can confirm we are on the same page:
1 - This field is populating under People in the view screen and not in the tab
Indeed, User/group picker custom fields are always displayed under the people section of the field, no matter which tab you add them. We reported the feature request below to implement the ability to change the location of these field types:

2. I'm not able to filter the issues with this field. Even though many tickets have this filed with a user value, I'm not able to filter it for a single user.
This is the default behavior of the Jira basic search drop-down in order to prevent performance issues when you have a big number of users on your site. Basically, it will only display any users when you type any values in the search field of the user picker, avoiding all the loading time to return all users at once:
We understand that this behavior can be a little cumbersome when the user does not remember the name of the user to be selected, so we also created a feature request to improve this behavior here:

This is really strange for me. I had added a custom user picker field few month ago and I did not have filtering issues with it and I still use that field for filtering issues. Here's a snapshot of it:
