Hannes from the JXL team here :)
You can't currently calculate values directly in JXL, although this feature (dubbed Formulas) is high up in our roadmap.
For now, what I would recommend is to use Jira Automation or an app like Scriptrunner to perform this calculation and write the result into a seperate field. This field can then be added to your JXL sheet, and will update if you change one of the "input fields". Many of our customers use this integration successfully, and we also use it internally.
