Hello!
I have imported some Requirements in Jira and need to update several values in the requirements based on Requirement ID.
I have a CSV file with the Requirement ID and the updated values.
My question is if it is possible to update the existing requirements using the CSV file and the Requirement ID as a matching key?

You would still need to map it to the issue key that has the Requirements information. But you should be able to do that, yes.

Just include a column in your CSV for the issue key and map it to the Jira issue key.
