How can I make the Work Log a mandatory field and the issue to require this information when there is a transition from In progress to another column (QA testing, Resolved, or Done)?
Also, how can I make the Original estimate a mandatory field and every task/ story to require this information when there is a transition from To Do to In Progress column?

May I suggest using a checklist app for this task? Smart Checklist, for example, allows you to add checklists to your Jira issues. You can make certain items mandatory, ensuring that an issue doesn't move to another status unless the mandatory checklist item is marked as done.
Alternatively, you can tweak your workflow to block transitions using the "Field Required Validator.
Go to issues > Workflows > Three dots to edit the workflow you'd like to tweak from the list > Click on the transition you'd like to edit > Select validators from the pop-up menu > Add the Field Required validator and specify the field.

Sorry, haven't noticed you are working on a Team-managed project. In this case the flow will be slightly different:
1. Click on the three dots in the right upper corner on the board
2. Select Manage workflow
3. Click on the transition that needs to be updated
4. Click on the "+" for the Rules on the right side
5. Select the following from the Rule list: Restrict to when the field is a specific value
6. Select the field you need and configure the remaining values
7. Click Save
8. Update the workflow

I don't have the option to select the field for Original Estimate. You can see below the available fields for me. Can you please advise on what is the reason for this?

Are you working with a Team Managed project or a Company Managed project? That information will show at the bottom of the panel on the left when you are viewing the project's board.

You can use the "Restrict (transition) to when a field has a specific value", as suggested by @Oleksandr Siryi_Railsware_ , for checking that the Original Estimate field is not empty.
Regarding requiring the Work Log, are you trying to (1) require the users to log work as part of the transition, or do you simply want to (2) confirm that some work has been logged against the issue at some point prior to attempting the transition?
(1) is not possible in Team Managed projects because they do not support adding a Screen during the transition to force entry of data while the transition is occuring.
(2) is possible by checking the Time Spent field in the same way you can check the Original Estimate field.

I apologize. I spoke without first validating my suggestion.
It does appear that neither the Original Estimate nor the Time Spent field is available for selection in that workflow transition rule in Team Managed projects.
I have not yet found any documentation or change request that specifies what system and custom fields are available, but I am continuing to look.
Given that you are on a Standard plan, you could also reach out to Atlassian Support directly to see if they can find and provide that information more quickly.
