Setup default configurations of columns for Kanban board

Default configuration of Kanban board comes with just three columns. We have added columns via the Tools > Configure > Column. But it does not reflect in the board. Some problem.

Also where can we setup a default template with additional columns so that all boards inherit it ?

There's no way to do this at the moment. We'd suggest just keeping one board as your 'template' board then copying it.
