For the project, I set a unique key.

I really need the suffix (number) to be specified not automatically, but in manual mode.

Could you define what you mean by "configure" and "flags"? Jira itself offers various fields that can be used to manage issues in a project and globally. Labels, components, versions, different kind of custom fields etc.. But is this question connected to the first one with the issue numbers? sorry to break it to you but you can't influence that issues will be consecutively numbered (unless you reset the counter in the DB) when they are created. This is set in stone and so you should spend time on what workaround in issues is a viable solution for you and the users (so prefixes or suffixes within the summary, custom fields etc.).
