Update labels or custom fields on create without using plugins

I need to create all tickets in a project with a label or a value in a custom field. Can i do it without the help of plugins ?

You can add that custom field in the "create screen" of the issue and make that custom field mandatory. This is built-in functionality and doesn't need any plugins.
