I would like to kindly ask if there is a way how to automatically assign to a First Tester or a First Developer using Post Function in the Workflow without using Plugins. Server based JIRA.

@Jack Brickey, by "first" I mean the first tester / developer assigned to. This might be set in a Custom Field called accordingly or something. I checked the Automation Lite Plugin, but it seems the functionality what I am looking for is a part of the paid Pro version. I would rather not used any Plugins for I am not sure about the support in the future and I would rather avoid changing the whole project workflow because some Plugin stopped being supported.
Anyway, I would like to have most of our simple processes in JIRA automated eventually. For example:
- issue is created with a Custom Field called First Tester filled, once developer finishes it and moves to the Done status, the transition to it changes the Assignee to the First Tester automatically on Post Function
- issue is reopened, the transition to Reopened status changes the Assignee to the Last Assignee (the developer who switched it to Done) or it could be handled the same way as with the First Tester above (that the dev would be assigned on issue creation in a Custom Field and then changed to) or something
The rest of the stuff I managed to get automated, but these two are the only problems remaining and I am sure it would save us a lot of time.
I am very new to the JIRA administration, thus the strange questions, but changes are needed in our company, so I am trying to make it somewhat better.
If you think this can be done I would kindly ask for a few steps how to.

you should provide more details on your requirements so that we could definitively answer. What does "first" mean here?
Maybe the use of Components could work? Or you mention "without plugins", do you not want to use plugins or you just don't want the up-charge. If the later then consider the free Automation Lite for Jira plugin.
