when I have added user to a specific role, when I do automation create one issue and it will automatically assigned to user in specified role. it was happened for the first time but when I create next issue and the issue would not assigned to the specified user in that role it was assigned to some one who not in the project role.
@Vishnu Vardhan Chennacheck whether the assigned user is in a group that is set up as default member for the role.

no there is no error message but the issue got assigned to some one who is not in the project role

So does the rule trigger and indicate success? If so then please check the History of the issue. I suspect someone or some other rule might be changing the assignee. Without seeing some of the details it is hard to troubleshoot.
