We are considering an Enterprise account for Trello, and the question has been asked as to what happens to cards, boards, and associated data when a user leaves the business. Can this be rolled up and reassigned to others, or does it get "lost"?

"Deactivating users removes the user's access from the team and the team's boards, but you're still able to see the cards to which the user belonged. This is helpful when trying to re-assign a member's work when one of your team's members leaves." It's best to re-assign the card and/or board before deactivating the other user.

One more thing, is it possible when transitioning people from free access to an Enterprise account, to do so in a way that their work is brought over and not lost?

When you transition to Enterprise, you will have to create a team. That team will consist of the individuals who would have access to the enterprise features. Then you will associate the boards with the new team. Therefore the work will not be lost.
