For some cards Im not able to add a checklist any more, any one an idea?

Hi @Gijs van der Stap , Welcome to the community!
From your screenshot I can see it's a new Trello's card back .Trello's card back was redesigned with the intent of improving the visibility of relevant information on the card.
To add a checklist item in this view, you need to click on the "+" icon and then select the checklist option.
As you mentioned you are not able to add checklist for specific cards ,so I have raised a support ticket here and I will ask more information on it to investigate the issue .
