Have 5000 user licenses for on premise DataCenter and want to move 30 of the users to offsite in the Amazon cloud .

No. The licence for a server (or cluster) belongs to that server.
If you want to have some people spin up their own installation somewhere else, they will need to licence that new server with a separate licence.
Can you not migrate existing licenses to the new server in amazon?

By "new server in Amazon", do you mean a separate Jira Data Center instance that's running in AWS (but set up by your IT staff)? Or do you mean a Jira Software instance in Atlassian Cloud, example yourcompany.atlassian.net?
The data center licenses are not splittable - they are all based on user tiers and tied to a specific instance as Nic mentioned. However the Cloud instances are billed per user per month and allow a little more flexibility since if you only need 30 users, you only pay for 30 users.
