We are not getting email notification when a ticket move from peer reviewer to manager approval.
Is there something specific we need to configured. We do get notification when the ticker first created

You can use the notification scheme as mr Goncalves mentioned above, but you can also use automation rules to send a notification to a person when a issue is being transitioned to a specific status. You can use the default automation plugin from JIRA but this is provided with limited options. I suggest you use "Automation for Jira" plugin:

Issues Created and Issue Updated notifications can be configured separated on the notification scheme of your project, so I recommend you to check if there is any missing group/role in the Issue Updated by navigating to Project settings > Notifications.
