Can Jira automation rule be used to make a specific custom field mandatory when transitioning an issue from one status to another?
For example if I transition my issue from 'Open' -> 'In Progress' I want the user to make sure Due Date has been populated.

Automation cannot be used to make fields mandatory. However, you can use automation to trigger post-transition actions such as adding a comment or sending an email etc if a field is left empty.
Requiring fields can be achievable via workflow validators.

For this task my suggestion is you use a screen to transition in a workflow with this field set as required or with a validator.

Automation can filled a field based on ruled, but it is not capable to create a behavior or define a field as required.
