If I have the following team setup how can I set up to automatically route tickets to the correct team leads?
Implementation
USA - Bob(Team lead), Jay, Ashley
MEXICO - Sara(Team lead), Kyle
CANADA - Mike(Team lead), Nick, Colin
Engineering
USA - Dolly(Team lead), Will
MEXICO - Phil(Team lead), Molly
CANADA - Connor(Team lead), Crystal
Do I set up a IMP and ENG project and then USA, Mexico, and Canada as labels?

If you want to keep the work that Engineering does separate from Implementation, then yes a project for each probably makes sense.
As far as automatically assigning to the Team Leads with Labels, I'd actually recommend that you look at Components instead:

Components will allow you to define a Component lead who can be the default assignee when the component is set, which sounds exactly like what you are looking to accomplish.
