How to Identify the most used plugins JIRA/Confluence. It is very difficult Identify based on the functionality of the plugin(suppose we have 50 plugins, based on functionality we need to check every plugin). Is there any way to Identify the list of most of used plugins In JIRA application?

Plugins work in such varying ways, you need a different rule for each type to determine a number for "most used", the number would mean different things for different add-ons, and many of them don't track usage at all.
Taking some very basic examples:
Define what you mean by "used" for a custom field type. Number of issues that have fields of that type filled in? Number of projects that have it available?
Define what you mean by "used" for a report add-on. Probably the number of people who use the report and how many times. But that's not recorded anywhere.
Define what you mean by "used" for a confluence macro add-on. This is about the only one that can be done, as you can search your whole system for the number of pages it is used on
Your only real option is to go through each add-on, have a look at what it does, define how you want to count "used" and then work out how to count that up.
