Anybody ever this kind of error and figured out why it was happening ? :

I cleared JIRA internal caches (using Script Runner built in function).
Also tried approving with different users.
Same results. On the same approval step.

I am able to reproduce this on a 4 approvals workflow). At the 4th, the error always occurres.

The error message that you received has to do - presumably - with either your Server (if you are hosting the service desk instance) or Atlassian's servers. All that the status code means is that there was some error server side. You can read a bit more about why that code might be returned in an HTTP request here.
