The catch is that this functionality is restricted for the JIRA Service Desk application and projects.
So, in a scenario where JIRA Service Desk is not available for you, you will need to rely on a plugin or something like that to perform this. Time to SLA is an example of plugin like this: https://marketplace.atlassian.com/apps/1211843/time-to-sla?hosting=server&tab=overview

yes you can. Easiest thing would be, to use the 'Duedate'.
The question is rather, what do you want Jira to do with a ticket that missed it's deadline?
