Hello, 

Is it possible to prevent user to close ticket if is not assign to anyone ?

You can set a condition on the transition in you workflow. By default you can use the "Only assignee condition". This will make sure that the user who want's to do the transition is the current assignee.

With the JSU Plugin you can also use the "Field Value Condition". With this condition you can make sure the the assignee field is not empty.
